I definitely asked for the number and setup too soon which probably put her off. I can get away with asking for the number on the second message ONLY because I run a sexual profile and mention casual short-term dating. If you are running a more traditional style profile/bio I can only imagine it will be tougher for you to number close on the second message.
I also stick to only using two emojis: the smirk one or the smiling devil, using these only when I send a polarizing message to soften it. Anything else comes off a bit feminine in my honest opinion.

Asking them out on the first message is probably too soon.
A lot of women don't feel comfortable giving out their number on the first message. They want to vet guys a little bit first. See if he's well-adjusted, not a weirdo or anything. I'd try to build some rapport with a few messages (like 5-10 messages?) before asking her number.
